I was checking the mailroot/Queue folder on my SMTP server and found quite a good number of .eml files struck in there.

For many messages in the Queue folder, eml file says -

'This is an automatically generated Delivery Status Notification.  
Unable to deliver message to the following recipients, due to being unable to 
connect successfully to the destination mail server.    

Though it's quite clear that it is because of the wrong email address with
hotmail.co instead of hotmail.co.uk

What can I do to remove this message from queue folder so that mail server doesn't keep on trying to send such messages with wrong email addresses?

Can you update the question with which mail server software is running? It could also be people didn't hit the m for .com. –  becomingwisest Sep 19 '11 at 16:17

That isn't helpful; we need to know which SMTP server: postfix, sendmail, exim, exchange, lotus notes, qmail, something else? –  MadHatter Sep 20 '11 at 9:34
Not very sure about which SMTP server :-( But, I think we are using SMTP server on IIS 6.0. –  iniki Sep 20 '11 at 10:48
