I have configured a postfix instance. But when try to send mail, I get this error:

And when I comment the relayhost parameter, mail is sent successfully.

How can I fix this?

In addition to NickW's question...It says, "connection timed out". Can you connect to port 25 on mta6.am0.yahoodns.net from the command line (using, e.g., telnet)? Is there a local firewall blocking outbound connections on port 25? –  larsks Mar 13 '13 at 13:18
I have checked and no firewall is blocking the port 25.And I also checked using telnet its successfully working on port 25.Further when I comment the relayhost line them mails are delivered successfully to my account. –  jenni Mar 14 '13 at 5:10
