Give the system some time to replicate the changes (and make sure the names.nsf is the same on both server). Depending on the config you have this might need a router reload (would prefer to reboot the domino server). Additional you could try a "Route *" on the console. It might also helpful to get a output from "Tell Router show". You might also try to route emails by hand as explained here until the issue is solved.

That hardly depends on your environment. Currently it seamed that the system isn´t able to find the needed SMTP Server. So if the internal routing is working please check if your outgoing server has a server connection document mentioned above. @rhsatrhs ist for outgoing emails, not internal ones as far as I understood the problem here.

I just tried these two Windows 2012 R2 SP1 Server. BUT on the first with desktop experience installed (the full bloatware) I have the option to perform many other features for example "Windows Update Cleanup", "Error Reportings" or "Logfiles". I do not have these options when I only copy the mentioned two files, so it looks like we miss some more then...

Yes I found that snipplet already. But its not working, if you will try to use -contains it means you NEED be directly added to that object. If you use AD groups, which is the case in most AD environments it will never work as you need to go true all groups at first. So this isn´t really the solution.

Additional to the above make sure you aren´t the one who enforce TLS and have a fallback in place. This can be configured via the Notes.ini entry RouterFallbackNonTLS

Is the resource DB replicated? See this link here. Make also sure that the replication works ... you can see that in the history. Make also sure that the RnRMgr task is running as mentioned in the URL above.
